In laser scanning microscopy using a high numerical aperture (NA) objective lens, spherical aberration caused by refractive-index mismatch in the specimen severely degrades focusing performance. This results in reduced signal intensity and degraded spatial resolution, particularly in deep regions. Here, we report the experimental results of aberration correction performed under conditions of significant refractive index mismatch, using a correction wavefront based on Zernike polynomial expansion with modified pupil coordinates, a method we previously proposed.
