Revealing Previously “Invisible” Molecules: Fixation and Immunostaining Strategies for Advanced Tissue Localization

Immunohistochemistry is an exceptionally versatile and powerful technique; however, insufficiently validated signals and overinterpretation remain persistent concerns in the literature. This lecture highlights key strategies for reliable cellular and subcellular localization analysis: (1) stringent evaluation of primary antibodies; (2) rigorous use of negative controls, including knockout tissues; (3) optimization of fixation and staining protocols, with a particular focus on glyoxal-based fixation and antigen retrieval; and (4) consistency assessments through correlation with mRNA distribution and comparative analysis across brain regions. Through sharing specific protocols and practical troubleshooting insights, I hope to discuss the essential considerations for generating localization data that are reliable, reproducible, and convincing.
