[2P-067]Prototype Screening of HaloTag-based Chemigenetic Indicators

Recently hybrid chemigenetic indicators have emerged as a promising new design. One representative is based on a protein structure with a protein-based sensing domain, while an environmentally sensitive synthetic fluorophore is fused to enhance fluorescence properties. However, to date this design hasn’t shown wide applicability to different analytes. Here, we explore the versatility of this framework based on the self-labeling HaloTag protein with a variable protein-based sensing domain, while a fluorogenic rhodamine derivative is fused as fluorophore. So far, we’ve designed two chemigenetic indicators based on this framework targeting Ca2+ and L-lactate respectively, and both showed promising fluorescence response.
