We introduce a dynamic optical coherence tomography (DOCT) method to assess the speed of the intratissue activities of in vitro samples. The DOCT method is based on the computation of time variance of the dB-scaled OCT intensity. An analysis of the acquisition-time dependency of the time variance provides two DOCT contrasts simultaneously. The relationship between each DOCT contrast and scatterer motion was numerically investigated. In addition, a human breast cancer spheroid was investigated with the DOCT method, which clarified the scatterer-motion properties in the sample.
