[P39]Limited tryptic digestion of hydrophilic domain of particulate methane monooxygenase

Particulate methane monooxygenase (pMMO), which converts methane to methanol, is embedded in the cell membrane of methanotrophs. Its protein structure is divided into a hydrophilic membrane-exposed domain and a hydrophobic membrane-bound domain, and copper ions, which are expected to be involved in the enzyme activity, are bound to both domains. In this study, the hydrophilic domain of pMMO was hydrolyzed with trypsin to investigate the effect on the enzyme activity. As a result, it was found that the catalytic activity and stability of pMMO decreased due to the hydrolysis.
