Human milk oligosaccharides (HMOs) play important roles in the brain development and health maintenance for infants, which are the third abundant components in the human milk. N-Acetyllactosamine (LacNAc) is an important intermediate for biosynthesis of other essential HMOs and antigens. Since the current synthetic methods including chemical and enzymatic ones are not appropriate for large-scale production of LacNAc, it is urgently needed to develop an efficient and cost-effective synthetic pathway of LacNAc. We established a one-pot multi-enzyme system to biosynthesize LacNAc and optimized the system. Finally, the LacNAc yield could be up to 90.5% under the low-cost condition, which would possibly meet the requirements of potential industrial production of LacNAc, and would provide a guide for production of other HMOs or functional oligosaccharides in the future.
