In this study, we propose a method to evaluate and correct spherical aberration in laser scanning microscopy by observing the axial information of PSFs from a single image acquisition using multiplexed focal spots produced by wavefront control of an excitation light beam. This method enables the prompt preparation of a dark spot applicable to STED microscopy even under the presence of spherical aberration in high NA focusing conditions.
