Fluorescence methods using enhanced electric fields by surface plasmon resonance (SPR) are one of the highly sensitive detection methods. However, the preparation of fluorescently labeled molecules is difficult and measurement process is complex. We have developed a dark-field microscopy using a grating-coupled SPR to enhance scattered light and enhanced scattered light from nanoparticles with a diameter of 200 nm was confirmed in our previous report. In this study, we evaluate the enhancement effect of the scattered light from even smaller nanoparticles and aim for label-free and highly sensitive detection.
