The liquid-ordered-phase (Lo-phase) domain can be stabilized by decreasing the thickness of the thermal oxide film that forms an overhang at the microwell apertures. However, the mechanism is still controversial. In this study, we investigated the relationship between the microwell aperture geometry and Lo-phase-domain stabilization. We concluded that deformation due to thinning of the overhangs causes changes in the distribution of curvature and stress in the freestanding membrane over a microwell, resulting in the localization of the Lo-phase-domain.
