A more detailed understanding of the mechanisms of lipid uptake and accumulation in hepatocytes is important to elucidate the disease mechanisms of MASLD and to achieve a very early diagnosis at a reversible and treatable stage. In this study, we investigated the pathological role of lipid accumulation in hepatocytes by Raman analysis of HepG2 cells cultured in a mixed fatty acid/cholesterol culture medium. We found that the degree of unsaturation, an index to assess lipid uptake, decreased with increasing cholesterol concentration in the culture medium.
