We propose a new imaging method for quantifying oxidative and reduced cytochrome c oxidase in the rat brain in vivo. This method analyzes the second derivative of the attenuation spectrum between 600 and 650 nm to estimate the absolute values of oxidative cytochrome c oxidase and reduced cytochrome c oxidase. A Monte Carlo simulation of light transport in a rat brain model is carried out to numerically establish the empirical formulae. In vivo experiments using an exposed rat brain while changing the fraction of inspired oxygen confirm the feasibility of using the proposed method to quantitatively image the redox state of cytochrome c oxidase in rat cerebral tissues.
