To elucidate the pathogenesis of metabolic dysfunction-associated steatohepatitis (MASLD), we analyzed the molecular characteristics of lipid droplets. We introduced a mixed medium containing fatty acids and cholesterol into HepG2 cells and evaluated lipid dynamics and metabolic processes after lipid droplet formation using Raman spectroscopy. The results demonstrated that the degree of unsaturation of fatty acids within lipid droplets varied depending on culture conditions and time, suggesting that the molecular characteristics of lipid droplets may serve as an indicator reflecting intracellular lipid metabolism.
