We investigated optical trapping at the surface of a protein solution and showed the formed assembly undergoes surface deformation characteristic of aged protein solutions. We analyzed the surface height change by monitoring transmission images of added PS MPs at the solution surface and estimated the surface deformation process. Before optical trapping, the surface of the saturated lysozyme solution is nearly flat. Upon laser irradiation, a circular valley-like dent is formed around the focal point, while the surface height at the focus remains nearly constant. Upon switching off the trapping laser, the elevated central region disappeared, the dent reappeared, and the surface gradually returned to a relatively flat state. Such behavior will be considered not only from optical and photophysical perspectives but also in terms of the mechanical properties of soft material.
