Single-cell mechanotyping is crucial for diagnostics, yet standardization remains challenging due to signal variability. We developed a dual-constriction microfluidic device to validate ion current-based measurements of cell deformation, volume, surface charge, and relative mass. By integrating high-speed optical observation and multi-physics simulations, we clarified the correlation between ion current pulses and cellular dynamics. This platform provides a fundamental technology for establishing standardized, high-throughput, and high-precision single-cell analysis.
