In this study, we developed a technology that integrates wide-field total internal reflection fluorescence illumination using an optical waveguide chip with real-time fluorescence immunoassay, enabling high-throughput and single-cell-resolution imaging of cellular secretion. Furthermore, by applying deconvolution that accounts for the binding dynamics of antibodies to the acquired secretion images, we successfully reconstructed instantaneous secretion images for individual cells, allowing detailed visualization of spatiotemporal secretion dynamics.
