Giant unilamellar vesicles, which are widely used as reaction fields for biomembrane mimicry, can be formed by various methods, including water-in-oil emulsion centrifugation method with organic solvents between lipid bilayers and electroformation method without organic solvents. In this study, we focused on protons, which have a higher membrane permeability coefficient than other ions and investigated the permeability of protons with and without organic solvents. The results showed that residual organic solvents significantly affected proton permeation and the membrane potential.
