[3D12]Functional Analysis of Sodium Ion Channels and Development of a Drug Screening System Using a Device for Applying Membrane-Horizontal Voltage

We developed an artificial cell membrane system applying a membrane-horizontal voltage (VHORZ) to reactivate inactivated voltage-gated sodium channels. Since continuous measurement of these currents is difficult, we validated this system for drug screening using VHORZ -evoked currents.The device features Au electrodes sputtered onto a micro-apertured Teflon film with CYTOP insulation. Cell-free synthesized sodium channels were reconstituted into artificial membranes formed within the aperture.Without VHORZ, reconstituted channel openings were rare. However, applying VHORZ significantly increased continuous channel activity. Adding tetrodotoxin (TTX) during VHORZ application caused a clear concentration-dependent current reduction, confirming the activity originated from sodium channels. This demonstrates the VHORZ platform's great potential for discovering novel ion channel modulators.
