Doping of PEDOT (poly(3,4-ethylenedioxythiophene)) with pH indicator dyes provides a simple way to introduce pH sensitivity into electrochemical biosensors. This is because PEDOT can mediate the redox cycling of the conjugated system found in typical colour changing indicators. In this study, we investigate bromocresol purple (BCP) dye, which has established uses in spectrophotometric serum albumin assay, as a dopant to provide electrochemical sensitivity and specificity towards human serum albumin (HSA). Cyclic voltammograms of PEDOT:BCP electrodes exhibit expected ideal Nernstian pH response for two proton coupled electron transfer (i.e., 60 mV/pH). In physiological buffer conditions (1x phosphate buffered saline at pH 7.4), distributed oxidation waves shift anodically following incubation of HSA, indicating a local pH change consistent with the reported colour change mechanism upon dye-binding. This behaviour is promising for protein determination via electrochemical means.
