In biology, cells communicate with functional molecules within the cytosol. The cytosol of any cell constitutes a complex solution with properties that dynamically change by life's processes. Achieving molecular extraction from cells without the loss of their functionalities remains a significant challenge in the field of cell applications.
Here we develop a novel pressure-controlled delivery system using gold nanotubes membrane-based injectors [1-2] to control cytosol extraction and effective substance transfer between source/target cells. By controlling key parameters such as insertion time, buffer volume, and needle diameter, and monitoring changes in fluorescence intensity of dyes, we demonstrate the ability to extract the cytosol from source cells remaining at high viability. The extracted cytosols were around a few hundred femto volume, so we controlled the inner pressure to hold the extracted cytosol within nanotubes, inserted it into target cells, and then increased the inner pressure to deliver the cytosol into target cells, thereby enhancing intercellular communication and molecular exchange efficiency. Importantly, this system ensures a recipient cell survival rate exceeding 95%, indicating minimal cell damage during the transfer process. This pressure-controlled delivery system provides a versatile platform for studying intercellular communication, targeted drug delivery, and genetic material transfer.
