[3H05]Microbial effects on dissolved organic matters in deep groundwater

Microbial culture tests were conducted in deep groundwater, which were then analyzed by microbial community evaluation via 16S rRNA analysis and characterization of organic matters, to evaluate effects on dissolved organic matter (DOM) by microbial activities. The microbe culture test Samples were taken at 0 days and 3 months, followed by filtration. Microbial DNA was extracted from the filter papers and amplified by PCR, followed by sequencing analysis to determine the DNA sequence. Based on this, microbiome analysis was carried out to evaluate the microflora. The molecular compositions of groundwater DOM affected by microbial activities was determined by high-resolution mass spectrometry and the complexation ability of DOM with an analogous element of radionuclides was evaluated by the fluorescence analysis.
