【Purpose】 Sodium chloride (NaCl) has been reported to influence allergic diseases. Recently, Epidermal Sensitization (ES) has gained attention as a major route of sensitization in food allergies, involving skin barrier disruption and inflammation. However, the impact of NaCl on ES is unclear. This study investigated NaCl’s effect on ES using a mouse model and ovalbumin (OVA), a major egg allergen. Two conditions were tested: (1) co-application of NaCl and OVA, and (2) OVA application three hours after NaCl treatment.
【Methods】 Six-week-old female BALB/c mice were used. In Experiment(1), mice were divided into OVA solution in pure water and OVA solution in 5% NaCl groups. In Experiment(2), pure water or NaCl was applied, followed three hours later by pure water or OVA solution in pure water. In both cases, skin barrier disruption was induced weekly by tape stripping and 5% SDS. Serum OVA-specific antibodies were measured using ELISA. TSLP and TARC levels in skin were also assessed.
【Results】In Experiment(1), an OVA solid-phase ELISA was performed using serum collected in the third week of treatment. Both OVA-specific IgG1 and IgE levels were significantly lower in the salt water group compared to the pure water group. In Experiment(2), similar results were observed when the skin was not washed three hours after the application of pure water or salt water.We are currently conducting follow-up experiments in which the skin is washed three hours after treatment with pure water or salt water, in order to investigate the mechanism by which NaCl suppresses Epidermal sensitization.
