[Purpose] Sebum secreted from sebaceous glands mixes with sweat to form a protective sebum film that helps maintain skin health. Reduced sebum production weakens the skin’s protective function, potentially leading to severe itching and inflammation. Therefore, it is desirable to combine ingredients that promote sebum secretion with moisturizers. Seed watermelon (Citrullus mucosospermus (Fursa)) is similar to wild watermelon, exhibits moisturizing and antioxidant effects and is used in cosmetics and supplements. To determine whether seed watermelon could serve as a cosmetic ingredient, we investigated its effect on sebum synthesis.
[Methods] Seed watermelon juice was subjected to silica gel chromatography, and its components were purified and identified using sebum synthesis activity as an indicator. Cytotoxicity was performed using the WST-1 assay, and sebum synthesis was evaluated by Oil Red O staining (540 nm) of lipid droplets produced in SZ95 cells stimulated with linoleic acid (LA). To further analyze lipid synthesis-promoting effects observed in cells treated with coniferyl alcohol (CA), a comprehensive lipidomic analysis was conducted.
[Results] Methanol eluates (0–100%) of seed watermelon juice were tested for their ability to promote sebum synthesis in LA-stimulated SZ95 cells. In this study, we focused on the 20% methanol eluate and isolated and identified four compounds: CA, coniferin, isoconiferroside, and p-coumaryl alcohol. Among these, CA promoted LA-stimulated lipogenesis by approximately 1.7-fold at a concentration of 300 µM. Lipidomic analysis further confirmed an increase in total lipid content following CA treatment, with notable increases in cholesterol esters, cardiolipin, and simple lipids.
